產品介紹 Streptavidin is a tetrameric protein composed of identic subunits. Each subunit binds one biotin molecule with a KD of ~1x10-15M. The preparation contains an N- and C-terminal shortened variant (core streptavidin) with improved properties concerning homogeneity, solubility, resistance towards proteolytic degradation and accessibility of the biotin binding pocket as compared to native streptavidin. The high affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits.


鏈霉親和素(Streptavidin,SA)產品簡介
鏈霉親和素具有與親和素(Avidin,AV)相似的生物學特性,如都能與4分子的生物素(Biotin)特異性結合,結合常數為1×10-15/M。同時,由于SA不帶糖基、等電點低,在檢測中具有比AV低的背景而大大提高了檢測的靈敏性。
生物素與親和素/鏈霉親和素的結合具有高度特異性和穩定性,兩者的親和常數比抗原-抗體反應高10-100萬倍,是目前已知強度最高的非共價作用,這使得它成為免疫學檢測中的一個非常好信號放大的工具。廣泛應用于酶聯免疫反應、熒光分子標記免疫反應和分子雜交為原理的各種檢測試劑盒的SA-biotin系統。但目前,國內所用SA均為價格昂貴的進口產品,因此研制和生產國產化的SA具有重要的現實意義和廣闊的應用前景。
????鏈霉親和素是與親和素具有相似性質的一種蛋白質,都具有與生物素特異結合的能力,結合常數高達1015M。同時,由于鏈霉親和素等電點比親和素低,且不含糖基鏈,故在檢測中的靈敏度和特異性都高于親和素,建立在此基礎上的生物素-鏈霉親和素系統的應用比生物素-親和素系統有更大優勢。
????我公司生產用鏈霉親合素-SA菌株及生產工藝均自國外原本帶回。生產工藝穩定,保證質量,常年提供產品,確保供應。
我公司生產的SA單體經SDS-PAGE檢驗分子量為17.2KDa。經論證,完整的SA溶解性差、易于聚集,核心SA被蛋白酶水解掉了與活性無關的某些末端序列,一方面提高了結構的穩定性;另一方面由于減少了空間位阻,使SA更容易與生物素標記的大分子發生反應。
SA的純化是用2-亞氨基生物素瓊脂糖珠4B 凝膠(Sigma 產品)進行親和層析純化,同時以離子交換進行進一步的精細純化。
SA分子量為58KDa,生物活性15.7U/mg,接近理論值(16.8U/mg)并進行了活性測定。
SA溶解與保存:我公司提供的SA產品,不含任何鹽離子,用戶可以根據自己的試驗需要,使用不同的緩沖液進行溶解,溶解后應放與-20°C保存,避免反復凍溶。
